The sleeve that came with this glass negative labels the sitter Robert E. Lee, but that title is unverified, so the identification should be read with caution. If the date of 1860 holds, Lee was then a colonel in the United States Army, three years before he took command of the Army of Northern Virginia and led it into the fighting at Gettysburg in 1863. The image is a wet collodion negative made by David B. Woodbury, who died in 1866. That process required photographers to coat and expose each plate while it was still wet, and the collection it belongs to is now held by the Library of Congress.
